For best DVD Audio Commentary, I nominate the commentary for the 2001: A Space Odyssey 2-Disc Special Edition, featuring Keir Dullea and Gary Lockwood.

It's not a perfect commentary - Lockwood gets some facts wrong and rambles at times, plus some other quibbles - the biggest complaint being that they didn't get the two men together in the same room for the commentary (unless I'm wrong, it feels like two separate commentaries edited together).

But...Lockwood's opinions and bluntness with some his comments is refreshing and humorous. I find Dullea to be the more informative and well-spoken of the two, not to mention that Dullea has a voice I find I could listen to for hours. Plus, I did learn some things I hadn't known before (possibly because I have yet to view the additional supplemental material).

What it boils down to is, the combination of Dullea and Lockwood kept me interested for 2 1/2 hours - and that's quite a feat, considering that I had just viewed the film in its entirety just the previous week.
